Throughout New York, many stretches of the byway have bicycle lanes and pedestrian access. This accommodation varies from city to city, so use discretion when walking or biking along the way. The eastern section is known to be a good bicycling spot.

The Great Lakes Seaway Trail in Pennsylvania features paved shoulders throughout on Route 5 for biking. The Bayfront Bikeway, a continuous paved multi-use trail system, will soon span the entire Bayfront.
